Israeli military closed the pathway leading to Naharayim enclave on Saturday ahead of the territory reverting to Jordanian control.
Israeli citizens and tourists can no longer visit the location.
Jordan announced its intention to cancel the 25-year renewable lease of Naharayim in October 2018.
